CULTIVO IN VITRO DE PLÂNTULAS DE ORQUÍDEA EM MEIO DE CULTURA ALTERNATIVO

Abstract

A micropropagação in vitro de orquídeas a partir de sementes é uma alternativa viável para a obtenção de mudas em condições controladas, visando o abastecimento do mercado e a preservação de espécies ameaçadas de extinção. Os meios de cultura tradicionais são complexos e de custos elevados, dificultando o acesso aos pequenos produtores. Este trabalho teve como objetivo avaliar a eficiência de um meio de cultura alternativo no desenvolvimento de plântulas de Brassavola tuberculata (Orchidaceae), comparando-o com meios tradicionais. As sementes de B. tuberculata foram submetidas a três tratamentos: T1 (meio Murashige & Skoog); T2 (meio Knudson C); T3 (meio alternativo). Foram retiradas 30 plântulas em cada tratamento para pesagem de massa fresca aos 145, 175 e 235 dias após a semeadura. Os dados obtidos foram submetidos à análise de variância e ao teste de Tukey. Em T1 as plântulas apresentaram crescimento menor nas três pesagens e, em T3, maior crescimento. A maior diferença entre os meios foi observada na terceira pesagem (p < 0,05) entre (T1 e T2) e (p < 0,01) entre (T1 e T3) e (T2 e T3). O meio de cultura alternativo (T3) promoveu o melhor desenvolvimento de plântulas de B. tuberculata.   Palavras-Chave: Orchidaceae; Brassavola tuberculata; micropropagação; preservação; alternativa de cultivo.     ABSTRACT    The orchid’s in vitro micropropagation from seeds is a viable alternative for obtaining seedlings in controlled conditions, aiming to market supply and preservation of endangered species. The culture mediums traditionally used are complex and expensive, difficulting the access by small producers. This study aimed to evaluate the efficiency of an alternative culture medium to the seedlings development of Brassavola tuberculata (Orchidaceae), comparing with traditional mediums. The seeds of B. tuberculata were subjected to three treatments: T1 (medium Murashige & Skoog), T2 (medium Knudson C); T3 (alternative medium). A total of 30 seedlings were removed in each treatment and the fresh mass weighted at intervals of 145, 175 and 235 days after sowing. The data were submitted to the analysis of variance and Tukey test. In T1 the seedlings showed lower growth in the three weightings and, in T3, higher growth. The greater difference among the mediums was observed in the third weighting (p <0.05) between (T1 and T2) and (p < 0,01) between (T1 and T3) and (T2 and T3). The alternative culture medium (T3) promoted the best development of seedlings of B. tuberculata.   Key Words: Orchidaceae; Brassavola tuberculata; micropropagation; preservation; alternative cultive.
